Synthèse de nouveaux analogues de la granulatimide à visée antitumorale

Many important biology molecular researches highlight the existence of cellular cycle regulator. They could operate in the development of the cellular cycle doing so as control quality. When anomaly is detected, as DNA damage, they could stop his progression and allowed DNA repairs. They are anymore two cycle cellular checkpoint in G1/S and G2/M. Recently two compounds have been isolated by an ascidie, Didemnum granulatum, the granulatimide and isogranulatimide. Theirs compounds have an interesting inhibitor activity toward the G2 checkpoint. Indeed, most cancerous cells have a deficient G1 checkpoint consequence of mutation of the p53 gene and so only the G2 checkpoint allowed the cycle cellular arrest. It result that the simultaneous action of inhibitors of the G2 checkpoint and DNA damaging agent may along early and lethal mitosis. In this work, the first part is devoting to the presentation of the cycle cellular G2 checkpoint and most particularly the checkpoint one kinase and her inhibitors. In a second part, we introduce synthesis of granulatimide's analogue where the imidazole heterocycle have been replaced by maleimide. A lot of synthesis pathway have been developed. We also carry out the synthesis in 7 azaindole series. In a third part the antiproliferative activities was exposed. All the compounds were evaluated by the SERVIER Laboratories.
